Overview

Alzheimer's disease research is the multidisciplinary scientific effort to understand the causes, progression, diagnosis, and treatment of Alzheimer's disease, the most common neurodegenerative cause of dementia. The field investigates the pathological hallmarks of the disorder, including the extracellular accumulation of amyloid-beta plaques and the intracellular aggregation of hyperphosphorylated tau into neurofibrillary tangles, alongside synaptic loss, neuroinflammation, and progressive cortical atrophy. A major emphasis is early detection, since neurodegeneration begins years before overt cognitive decline; researchers evaluate biomarkers in cerebrospinal fluid and blood, structural and functional brain imaging, and emerging approaches such as retinal optical coherence tomography to identify preclinical change. Cognitive, behavioral, and sleep disturbances are studied both as clinical features and as potential modifiable contributors to disease course. Therapeutic research spans pharmacological strategies aimed at amyloid, tau, and inflammatory pathways, as well as non-pharmacological interventions for sleep disruption and behavioral symptoms in moderate to severe dementia. Investigators also examine neuromodulation, including vagus nerve stimulation, as candidate approaches. Across these efforts, the discipline relies on longitudinal cohorts, animal and cellular models, and systematic synthesis of evidence to translate mechanistic insight into improved diagnosis, prevention, and care.
