Overview
Necrosis is a form of cell death resulting from acute injury, in which cells lose membrane integrity, swell, and rupture, releasing intracellular contents that provoke inflammation and surrounding tissue damage. Unlike the regulated, non-inflammatory process of apoptosis, classical necrosis is typically uncontrolled and follows ischemia, infection, toxic exposure, or physical trauma, though regulated necrotic pathways are also recognized. It is implicated across infectious, neoplastic, ischemic, and degenerative disease and contributes to organ dysfunction through tissue destruction and the release of damage-associated molecules. The peer-reviewed research collected here reflects these themes through reports of lymphoma with extensive necrosis, quercetin attenuation of manganese neurotoxicity involving tumour necrosis factor alpha regulation, and reversible and irreversible cerebral necrosis following cardiorespiratory arrest. Related contributions examine apoptosis and oxidative stress in stored platelets and in testicular tissue under lipopolysaccharide challenge, computed-tomography severity indices and biochemical markers in acute pancreatitis, and proinflammatory cytokine expression in tissue homogenates. Further work addresses neoplastic tissue damage, adrenal structural change under chemical exposure, and oxidative end products in hemodialysis. Methodologically, the literature draws on histopathology, biochemical and cytokine analysis, animal models, and clinical case study, illustrating how necrosis connects cellular injury, inflammation, and tissue destruction across infectious, toxic, ischemic, and neoplastic conditions, and how its assessment informs understanding of disease severity and progression.
