Overview

Vesicles are small, membrane-bound sacs enclosed by a lipid bilayer that compartmentalize, store, and transport molecular cargo within and between cells. They are central to intracellular trafficking, mediating movement of proteins, lipids, and other biomolecules among the endoplasmic reticulum, Golgi apparatus, endosomes, lysosomes, and the plasma membrane. Vesicles are classified by origin and function: transport vesicles coated by COPI, COPII, or clathrin mediate secretory and endocytic pathways; secretory vesicles store hormones and neurotransmitters for regulated exocytosis; and synaptic vesicles release neurotransmitters at chemical synapses. Their formation, budding, tethering, and fusion are governed by coat proteins, small GTPases such as the Rab and Arf families, SNARE complexes, and dynamin-dependent scission. In endocrinology, secretory vesicles underpin the controlled release of peptide and amine hormones from endocrine cells in response to physiological stimuli, linking vesicular dynamics to hormonal signaling and homeostasis. Extracellular vesicles, including exosomes and microvesicles, additionally carry proteins and nucleic acids that mediate intercellular communication and have growing significance as biomarkers and therapeutic carriers. Vesicle biology is fundamental to understanding membrane dynamics, cellular signaling, hormone secretion, and the development of targeted drug-delivery systems, and remains an active area of cell-biological and biomedical research.
