The Effects of L Carnitine on in Vitro Maturation of Immature Bovine Oocytes

L-Carnitine (Lc) acts as an antioxidant that neutralizes free radicals, especially superoxide anions and protects cells against oxidative damage-induced apoptosis, as following ovulation, intracellular reactive oxygen species (ROS) accumulation increases in oocytes, Oocytes exhibit an intracellular defense mechanism against an oxidative attack. This outcome adversely affects fertilization and subsequent embryonic development, thereby increasing the risk of an early miscarriage and abnormal development of offspring. The purpose of this study was to see how adding LC to either maturation or fertilization medium affected the developmental competence of immature bovine oocytes. In this study, Ovaries from apparently normal reproductive organs of cattle were collected within 30 minutes from slaughter and evisceration of animals. Cumulus oocyte complexes (COCs) were collected by aspiration of medium sized ovarian follicles (4-8 mm). COCs of acceptable quality were selected, washed and incubated in tissue culture media 199 (TCM199) supplemented with 10% heat inactivated fetal calf serum, 5 μg/ml luteinizing hormone (LH), 0.5 μg/ml follicle stimulating hormone (FSH) and 1 μg/ml estradiol-17β for 20:22 hour at 38.5 C◦ under 5% CO2 in air with 90% humidity. different concentrations of LC (1.25,2.5 and 5mM) were used. The results were consistent for both maturation and fertilization and there is a significant increase in maturation, fertilization., cleavage and blastocyst rate. In conclusion, LC has important role in IVEP through addition of LC to maturation media or culture media it improved nuclear maturation and blastocyst formation rates in bovine oocytes.

Dragonhead Cyclone in The Falkland Islands (Malvines Islands) has Double Cote's Spiral Like NGC 5247

Extratropical cyclones are common in the South Atlantic. They generally arise with the passage of cold fronts to the south of the South American continent, crossing the south of Chile and Argentina, in the regions of Puerto Natales, Punta Arenas (Chile), Rio Galegos, Rio Grande, El Calafate and Ushuaia (Argentina). The extratropical cyclone analyzed presented at its peak the very characteristic shape of a Chinese dragon. Other cyclones in the form of a Cote's spiral curve are part of this analysis (Gobato et al., 2018-2023).  They present a mathematical form of a double Cotes Spiral curve. Here called the Dragonhead cyclone, indicate a structural similarity with spiral galaxies, especially NGC 5247, in the constellation Virgo. With an area of influence and a size of around 3,247 thousand km2 at its peak, it moved quickly in a west-northwest (WNW) direction, with an average speed of 76 km/h, with winds of 84 km/h at 100 km from the nucleus, as it passed north of the Falkland Islands (Malvinas Islands). The cyclone temperature during its trajectory varies from -45ºC to -50ºC at its edge, while at its core it varies between -10ºC to -25ºC. In the data collected (Gobato et al., 2018-2023), and analyzed from the Dragonhead cyclone, it is clear that all extratropical cyclones that appear south of the South American continent, below 40° latitude, have the shape of a spiral curve, like the spiral galaxy. Most of these are in the form of a double Cote’s spiral curve.

Prevalence and Economic Importance of Bovine Hydatidosis in Animal Slaughtered in Dalomana Municipal Abattoir Southeastern, Ethiopia

A cross-sectional study was conducted on bovine hydatidosis from May 2018 to November 2020 with the aims of finding its incidence and economic losses in bovine slaughtered for human consumption in Dalomana municipality abattoir. Usual ante mortem and post-mortem inspection was carried out on overall of 430 selected slaughtered cattle. From this total inspected, 82(19.1%) of them had harboured hydatid cyst (s) in one or more of their organ’s. Age and body condition as a risk factor were statistically significant as older zebus and medium were more infected (P<0.05, χ2= 14.597 and p value<0.05 χ2=9.68) respectively. Similarly, the geographical origin of the animals had significant contribution on hyadatid cyst distribution (P < 0.05, χ2 =12.724). Sex was statistically non-significant (P>0.05,χ2 =0.74) and have no difference. The single and multiple organs were infected by hydatid cysts. From the total of 282 hydatid cysts collected and characterized lung and livertook more percentage in harboring cysts, from this 137(48.58%) and 126(44.68%) cysts were observed in lung and liver respectively and 19(6.74%) are observed in other organ, such as spleen, kidney, and heart. The study indicated that annual economic failure due to direct organ condemnation and indirect carcass weight loss from the disease in Dalomana was estimated to be 105769.657 ETB. Hence, bovine hydatidosis is significantly a common disease in cattle, with severe public health fears and leads considerable servable and undetectable losses in bovine in Dalomana and its surroundings.

Vineyard Clusters Monitored by Means of Litterbag-NIRS and Foliar-NIRS Spectroscopic Methods

There is currently a lack of rapid indirect analysis methods for the assessment of the effects of soil microbiota on vine production. Fifteen clusters of two Nebbiolo and Erbaluce varieties were identified in five vineyards belonging to a cooperative of winemakers in North West Italy, according to the differences in the NDVI index, as monitored by the Crop Monitoring OES system. The vineyards were surveyed in 2019 and the experimental monitoring of 75 vines was conducted in 2020. The first indirect method (Litterbag-NIRS) involved examining hay litterbags with a smart SCiOTM device. The average litterbag-NIR spectra of the clusters, as far as the yield is concerned, were closely fitted with the measured production yield, with an R2 cross-validated value of 0.91 in the Nebbiolo vines and 0.67 in the Erbaluce vines. The results in yield were accounted for by considering a few dominant variables in both vines, namely the microbic respiration of the soil and the crude protein of the litterbag (positive), opposed to the soil NO3--N and litterbag ADF (negative). The pruning wood was also closely correlated to the litterbag spectra. A second rapid method, foliar pH coupled with the NIR spectroscopy of the leaves, was then performed. The overall results predicted from the foliar NIRS were 0.73 for yield and 0.79 for the Canopy Cover. However, the most interesting result concerned the yield regressions on the foliar pH, which were clearly negative in both vines and of a similar amount: -5.15 kg/pH in Nebbiolo (R2 0.68) and -5.63 kg/pH (R2 0.23) in Erbaluce. Litterbag-NIRS, which shows a high predictive capacity, and foliar pH - with or without foliar-NIRS - are indirect and frugal methods that can be recommended for a rational assessment of the microbial soil fertility of vineyards.

Coprological Examination of Ovine Fasciolosis in Horro District Community Based Sheep Breeding Program, Horro Guduru Wollega Zone, Western Ethiopia

A cross-sectional study was conducted from November 2017 to March 2018 to determine the prevalence of ovine fasciolosis in the Community Based Horro Sheep Breeding site at Horro district, Horro Guduru Wollega zone, Ethiopia. Coprological examination was performed on a total of 390 sheep at the study area. Age, sex, body condition score and peasant association were taken into consideration as rik factors. An overall prevalence of fasciolosis 45.4% 95% CI (40.41 50.36 was found on the basis of coprological examinations. The prevalence of fasciolosis findings according to PAs were (41.5%) in Laku Ingu and (48.2%) in Gitilo Dole. Statistical analysis of the prevalence among study site (PAs) showed insignificant difference P=0.102 (P>0.05), but significant difference (P<0.05) was observed between animal age groups (P=0.032, with a prevalence of 37.2% in young and 49.8% in adult) and body condition scores (P=0.001, with a prevalence of 57.9%, 43.5% and 29.9% in sheep with poor, medium and good body condition score, respectively. There was insignificance difference (P>0.05) in sex group of study animals in which the prevalence was 47.6% in female and 38.8% in male. Therefore further studies on the epidemiology, seasonal dynamic of the disease, the snail intermediate host and impacts of the infection in animal production with implementations of strategic intervention is necessary.

Sunspots are Correlated with Foliar pH in Grapevines

Foliar pH is a specific multifaceted parameter that is sensitive to a deficit in soil water and to temperature variations. It also represents a tool that can be used to rapidly phenotype the symbiosis induced in several crops by bio-fertilizers containing Arbuscular Mycorrhizal Fungi. Yearly decreases in foliar pH, which dropped from 3.73 in 2015 to 3.15 in 2017 and then stabilized at around 3.13, have been observed in an experimental vineyard near Torino (Italy) in six grapevine cultivars. In this paper, these curious, original results have been paired with the average sunspots of the 24th sun cycle, proximal to its endpoint. The paired values were highly correlated (r 0.95 P< 0.01), with close parabolic patterns. A lowering in foliar pH has been correlated with a modification of the leaf composition, as characterized by the higher hydration and reinforced wall. An increase in the circulating acidity of the plants has been hypothesized to interfere in a diminution in the general predisposition to block parasite attacks. From this perspective, the retrieval of several historic outbreaks and the long-term systematic monitoring of mud and Erwinia amylovora frequencies have suggested that the hypothesis that links the solar minima with dysfunctions of the plant-pest relationships cannot always be rejected. Cosmic influences pertaining to UV variations are poorly understood in plant physiopathology. Foliar pH appears to be a rapid and simple tool to unveil high-level mechanisms. It is this simple parameter that physiologists and geneticists, but also agronomists, are asked to consider.

Effect of Coinfection by Fasciola hepatica and Mycobacterium bovis on Bovine Tuberculosis Immunodiagnosis in an Enzootic Area Hidalgo State, Mexico.

Parasitic infection by the Fasciola hepatica (F. hepatica) promotes susceptibility towards other infections, such as Mycobacterium bovis. As consequence, could affect diagnostic tests for this disease. Hence, the objective of this study was to assess the impact of F. hepatica coinfection on the most commonly used immunodiagnostic bovine tuberculosis (bTB) tests in field conditions in an enzootic area for both diseases. Thus, from a dairy herd located in Hidalgo State, México, displaying a 59.2% and 28% prevalence of fascioliasis and bTB, respectively. Sixty-one cows were analyzed based on their response towards bTB immunodiagnostic tests, such as Single Intradermal Comparative Tuberculin Test (SICTT), gamma-interferon test (BOVIGAM) and enzyme-linked immunosorbent assay (ELISA), along with the assessment of the F. hepatica parasite load and serodiagnosis by ELISA. Three study groups were formed according to test results. Group 1: coinfected (n=22). Group 2: non-parasitized cows, and positive for bTB tests (n=13) and Group 3: parasitized cows without tuberculosis (n=26). In addition, a group of cows kept in fascioliasis - and tuberculosis-free zones were included (Group 4, n=10). A non-parametric Kruskal-Wallis test and a Dunn test were applied to analyze the results. In Group 1, significant differences were observed regarding IFN-γ production, but not for antibody levels to M. bovis or reactivity towards bovine PPD in relation Group 2. While, Groups 1 and 3 did not display difference in antibody levels against F. hepatica. Differences were observed regarding tuberculosis and Fasciola diagnostic tests when both coinfected and infected groups were compared to controls. It is concluded that F. hepatica coinfection in tuberculous animals studied, depressed the production of IFN-γ towards bovine PPD under in vitro conditions, but its reactivity to the SICTT not show to be altered.

A Study on Contagious Bovine Pleuropneumonia in Khartoum State, Sudan

The study was carried out to investigate the occurrence of contagious bovine pleuropneumonia CBPP in Khartoum state. One-hundred twenty-two pneumonic lung tissue samples were collected from different slaughterhouses (116 samples most of which from local breed cattle) and from the field (six samples from cross breed cattle). Two-hundred and fifty-seven serum samples were collected randomly from cattle in different areas of the state. Tissue samples were cultivated using the standard mycoplasma procedures. Mmm was isolated from three pneumonic lungs collected from the field while no isolates were recovered from slaughterhouse samples. Histopathological sections from the positive samples revealed the typical picture of the CBPP which include fibrinonecrotic pneumonia within filtration of inflammatory cells and fibrin and distention of interlobular septae. One hundred and eight out of 257 serum samples were found positive for antibodies against Mmmusing complement fixation test (CFT). Findings of this study confirmed the presence of CBPP in Khartoum state by the isolation and identification of the causative agent.

Spiritual Blessing Energy Treatment as an Approach for Boosting Growth and Yield of Bottle Gourd (Lagenaria siceraria L.)

The objective of this study was to improve the growth and yields of bottle gourd plants after exposure with spiritual blessing energy treatment (SBET) to the seeds and land. Twenty-one morphological traits of leaves, fruit, seed, and twenty-eight phenological parameters were employed to evaluate bottle gourd’s growth and yields after SBET. Results of bottle gourd showed a diversifiable characteristic of leaves, fruits, and seeds. Nine qualitative traits exhibited with distinct differences in the biofield-energy treatment group compared to the control. At harvest, growth-related phenological parameters such as plant vine length, number of branches, internodal length, and leaf width were significantly improved in the treatment group by 65.43% (p ≤ 0.001), 51.78% (p ≤ 0.01), 49.40% (p ≤ 0.001), and 40.82% (p ≤ 0.001), respectively, compared to the control. Additionally, yield-related parameters such as number of female flowers, number of fruits per vine, and fruit yield per hectare were significantly increased by 47.73% (p ≤ 0.05), 26.52% (p ≤ 0.001), and 38.99% (p ≤ 0.05), respectively, with respect to the control. With distinct qualitative variations and high statistical significance, these results suggest that SBET may serve as a potent non-traditional intervention for optimizing phenological development and agricultural output of bottle gourd.

Morphological Attributes and Yield of Momordica charantia (Bitter Gourd) Under the Application of Spiritual Blessing/Biofield Energy Treatment (BET)

Objective Bitter gourd (Momordica charantia L.) is a widely consumed vegetable. This study aimed to assess the impact of spiritual blessing (biofield) energy treatment (S/BET) known as the Trivedi Effect® on a plant’s growth and yield when applied to both seeds and soil. Methods Seeds were split into two groups: an untreated control group (CONBIGG) and a BET-group (BTBIGG). Soil texture was checked by hand, and mineral contents were measured using standard procedures. An experienced scholar noted plant morphological, phenological, and yield-related parameters. Results The S/BET group showed improvements in stem length, fruit colour, and seediness compared to the control. Other phenological parameters, such as vine length (38.78 %; p ≤ 0.001), branch number (54.03 %; p ≤ 0.01), inter-nodal length (42.94 %; p ≤ 0.001), fruit length (33.27 %; p ≤ 0.001), seed length (40.32 %; p ≤ 0.001), seeds per fruit (52.50 %; p ≤ 0.001), and fruits per vine (66.88 %; p ≤ 0.001), were significantly increased in the BTBIGG group compared to the CONBIGG. Yield per hectare rose by 61.31 % in the S/BET group compared to the control. Conclusion The study results suggest that the spiritual blessing/prayers/BET-Trivedi Effect® significantly improved morphological, phenological, and yield-related parameters compared to the control group. This study provides useful scientific research data and information for producing stable, high-quality bitter gourds.

RETRACTED: Leap into Fetal Surgery; In Utero Placental Mesenchymal Stem Cell Therapy, A Contemporary Approach to Treating Myelomeningocele

According to the U.S. Centers for Disease Control and Prevention, each year approximately 1,400 babies born in the United States have spina bifida. The disease manifests with the lack of skin and bone covering the caudal part of the spinal cord. The patient developing such a condition often develops lifelong impaired lower limb mobility accompanied by hydrocephalus, and urinary and bowel incontinence. The available interventions include prenatal and postnatal surgery to fuse the dura. Prenatal surgery performed before 26 weeks of gestation reduces the risk of death or the need for ventriculoperitoneal shunting. It also enhanced results on a comprehensive index for mental and motor function. When compared to postnatal surgery, prenatal surgery reduces the manifestation of several secondary outcomes, including the degree of hindbrain herniation seen in the Chiari II malformation. Stem cell therapy for MMC on animal models of chick, ovine, and rodents with reported cases 15/63, 15, and 136, respectively, using human Embryonic Stem Cells (hESCs), Neural Stem Cells (NSCs), Mesenchymal Stem Cells (MSCs) showed significant coverage of MMC defect and slight neurogenesis was also observed. With an understanding of medical literature about in-utero regenerative capacity, it is to be appreciated that placental stem cells surgically seeded within a biocompatible scaffold of the cell patches can play a part in alleviating the spinal cord manifestation associated with MMC. Documented animal studies show that incorporating Placental Mesenchymal Stem Cells in prenatal surgery has reported improved neurogenesis and lower limb mobility. In an ovine myelomeningocele model, the development of in-utero myelomeningocele repair with human Placental Mesenchymal Stem Cells seeded onto an extracellular matrix (PMSC-ECM) enhances motor findings. The clinical trial for the first stem cell therapy on human subjects known as the “CuRe Trial: Cellular Therapy for In Utero Repair of Myelomeningocele.” is expected to be finished by 2030. So far, the cases undergoing treatment have shown significant leg movement and a greater degree of bowel and urinary control. This FDA-approved clinical trial is envisioned to be the future of treating MMC.

Soluble Biobased Substances in soil or Salicylic Acid on leaves affect the foliar pH and soil biovariability of Grapes-as explained by the NIR Spectroscopy of Litterbags and Teabags

The purpose of the trial was to check the effects of two grapevine treatments on the plant activity and on the bioactivity and biovariability of the soil. An alkaline complex of Soluble Biobased Substances (SBS) was used in soil at 30 g per plant in a single solution. Salicylic Acid (SA) was used on leaves at 150 mg l-1 every two weeks at 50 ml plant. The plants were examined for their foliar pH and NIR spectra. The soil bioactivity was monitored by means of hay-Litterbag-NIRS (LBN) in combination with the Teabag Index (TBI), using rooibos and green tea that had been buried for 60 days. The evolution of the TBI presented here concerns the TBI-NIRS spectroscopic method used for discriminant analysis. A new algorithm was used to estimate the soil microbiome from the green Teabag spectra. The obtained results showed that the plants and the soil responded to the treatments. In fact, SBS, but not SA, lowered the leaf pH by 5%, an unexpected and original result. Both treatments increased the variability of the leaf composition, with a lower discrimination, based on the NIR spectra, from the Control (75%) to 44% (SA) and 38% (SBS). The TBI method, which is based on weights, was less efficient (67%) than the TBI-NIRS of rooibos (96%) or the LBN of hay (80%), but it was like the TBI-NIRS of green tea (74%). The LBN analyses indicated that the mycorrhizal index had increased by 8% in SA but had reduced by 7% in SBS, while both treatments reduced the activity of the microbes, which did not affect the soil respiration rate. The mineral N in the soil was substantially raised by about 11÷69%. The Taxa profiles showed marked deviations from the Control. Moreover, the SBS treatment reduced the Glomeromycota by 35%, which matched to the reduction in the mycorrhizal index. The most favored Bacteria from the treatments were Proteobateriaand Actinobacteria, with Mortierellomycota being the most penalized. Neither treatment affected the production, but both delayed the technological maturity by 9-11%, while the SBS retarded the phenolic maturity by about 18%. It has been concluded that a simple treatment of vines can affect the bioactivity in the leaves and berries as well as the biovariability of the soil.

Cytokine Expression in Peripheral Blood Mononuclear Cell Cultures Obtained from Cattle with Different Stages of Natural Mycobacterium bovis Infection

In bovine tuberculosis (bTB), cellular, humoral, or both types of immune responses have been observed. The purpose of this study was to examine the immune status of tuberculous cows based on the differential cytokine gene expression associated with Th1 (IFN-γ, IL-2), or Th2 (IL-4, IL-10) responses. Twenty-three (23) cows belonging to a dairy herd located in a rural region of the State of Hidalgo, México, were selected for the study. Single Intradermal Comparative Cervical Tuberculin (SICCT) Test, Interferon-Gamma (IFN-γ) Release Assay (BOVIGAM), and Enzyme-Linked Immunosorbent Assay (ELISA) were used for detection of cattle infected by M. bovis. Thirteen cows were positive to all the tests (Group 1); ten cows were positive only to ELISA (Group 2), and the remaining Group (Group 3, control) included cows negative to all the tests. Peripheral blood mononuclear cells (PBMC) from animals were in vitro stimulated by bovin purified protein derivative (PPD), avian PPD, and Concanavalin A (Con A) mitogen for 72h. Changes in the levels of expression of mRNA of the respective cytokines was measured by Reverse Transcription-Polymerase Chain Reaction (RT-PCR) using β-actin gene as internal control. In group 1, PPD bovis and Con A-stimulated cells exhibited high production of IFN-γ, IL-2 and IL-4, but not IL-10. In contrast, PPD avium-stimulated cells displayed a low production of cytokine transcripts. In group 2, cells showed a significant production of IL-10 in response to bovine PPD (P< 0.001). In the control group, a high production of IFN-γ and IL-2 was observed only in Con A-stimulated cells. Post-mortem examinations in animals of group 1 showed slight and medium lesions in lymph nodes, whereas in group 2, the lesions were more extensive. Results indicate differences on gene expression levels of cytokines considered to determine balance in Th1/Th2 response among the evaluated groups. In addition, high levels of antibodies against M. bovis and high IL-10 expression in PBMC together are indicators of progressive bTB when both tuberculin test and IFN-γ assay are negative in tuberculous anergic cattle. Inclusion of serology and IL-10 cytokine expression in in the diagnosis checklist improves detection of infected cattle to help control bovine tuberculosis.

Colored Anti-Hail Nets Modify the Ripening Parameters of Nebbiolo and a Smart NIRS can Predict the Polyphenol Features

In the cultivation of vines the risk of hail appears to be increasing with the ongoing climate change. The use of protective plastic nets is spreading, but there is little knowledge on the additional processing costs and on the phenological and qualitative consequences, moreover, as depending on different colors of the nets. Thus, a randomized trial was carried out in Nebbiolo, a wine of great aging, testing three plastic anti-hail nets colored in green, gray and black. Results showed that 24% more-time was necessary for the canopy management practices under the nets. The black nets advanced the ripening process, with a berry weight decrease of about 13%. A significant decrease in the seed number per berry was observed under the black nets (-45%), while a rise (+102%) was observed under the green and gray nets with a parallel increase in the pH of the juice (+13%) and in malic acid (+28%) under the green nets. The berry skin did not show any significant differences in polyphenol and anthocyanin profiles, while the plants that matured in the upper part of the vineyard showed higher level in the berry skin extractable flavan-reactive to vanillin, and total polyphenol. On the other hand, in the seeds grown under green nets an increase in the extractable polyphenol compounds was observed, sign of a delay in seed ripening, with a higher tannin polymerization ratio, preserving the malic acid, and decreasing the acidity of the berry. In conclusion, the use of colored green may be a useful tool against excessive microclimatic warming and / or irradiation. The field internal variability affects much more the ripening dynamics than the nets used. A second aim was to develop a smart NIR SCiOTM model for polyphenols and the results were in line with the favorable expectations, providing R2 predictions of about 0.74 from the skin and 0.81 from the seeds.

Background COVID-19 as an infectious disease, and deadly biological crisis, threatens the bio-psycho-social- spiritual health of the people. Spiritual health from the perspective of Islam, means having a sound heart, living in the present time with sense of peace, security, patience and gratitude, safe from the fear and anxiety of future, grief and regret for the past events. It affects other dimension of health. This study was conducted to investigate the spiritual health services in the face of the COVID-19 pandemic in a Muslim society. Methods This qualitative study was conducted from March 1, 2020 to the end of May 2020 in Tehran by using the Schwartz and Kim's hybrid model concept analysis with a deductive / inductive analysis approach, in three stages: 1- Theoretical review of religious and scientific evidence, 2- Field research 3- Final analysis. Data collection was done in hospitals and hospices by semi-structured interviews, taking notes, websites and social networks search. Data were analyzed by "Contractual Content Analysis Method". Results Despite the fact that spiritual health services (spiritual care and counseling) are not taught in Iranian universities, but in this biological crisis, spiritual health services, based on religious beliefs of health system employees were implemented. The Muslims’ belief in divine test, healing power of God, helping the people as highest worship, aroused spiritual awakening and enthusiasm in the health care team. Spiritual health services at prevention levels were provided with the aim of helping the patient, family and clients, in an inter-professional model based on the jurisprudential rules derived from the religious evidences (Verses and Hadiths), in line with holistic approach, community-based care, spiritual self-care, home-care, family participation. The involvement of non-specialists in the provision of medical services was prevented. According to, preserving the human dignity in Islam, Islamic rituals were performed by the treatment team and volunteer clerics for dying and dead people. Conclusion Considering the impact of religious spirituality on Muslims’ lifestyle and health behaviors, it seems that the use of Islamic health guidelines can improve the quality of health care services and help improve the spiritual health of people in biological crises.

Molecular Evolutionary Characteristics of the 2019 Novel Coronavirus (SARS-CoV-2) Contracted by Tunisian Citizens : Comparison and Relationship to Other Human and Animal Coronaviruses Based on Spike Glycoprotein-Coding Gene Sequences Analysis

In contributing to the initiative to address the COVID-19 pandemic and in order to enhance the knowledge on driving forces shaping the evolution of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) (isolated from Tunisian patients), a comparison in relation to other coronaviruses infecting humans (SARS-CoV-1, MERS-CoV, HCoV/229E, HCoV/NL63, HCoV/OC43, and HCoV/HKU1) as well as animals (SARS-CoVs in tiger, bats, civet, pangolin, bovine, and MERS-CoV in dromedary/camel), was conducted. In-depth analysis was carried out involving 115 sequences of spike glycoprotein-coding gene extracted from the international databases. Phylogeny inference allowed the reconstruction of a bifurcating tree where four distinct groups were delineated and at the same time, three animal accessions (SARS-CoV-2/tiger, MERS-CoV/camel, and SARS-CoV/bovine) shifted from the animal group and integrated the human coronaviruses clades. Nonetheless, in the presence of reticulate events such as recombination, networks described better the phylogenetic relationships rather than the classic dendrogram. Thus, networks were produced and identified four clusters containing sharply demarcated subgroups (eight subdivisions). Except networked phylogenies of SARS-CoV-1, SARS-CoV-2, and HCoV/HKU1, all the others showed edges and boxes illustrating the occurrence of incompatibilities related to the sequences of spike glycoprotein-coding gene. Thereby and consolidating this result, three methods (RDP package, GARD, and RECCO) were used to detect breakpoints in aligned sequences. Except the clades SARS-CoV-1 and SARS-CoV-2, all the remaining phylogenetic subdivisions were subject to recombination. Furthermore, the screening of selection pressure in all studied sequences by various statistics-based models of the HyPhy package, showed that, similarly, the lineages belonging to the clades SARS-CoV-1 and SARS-CoV-2 were not under selection. In contrast, all members of the remaining clades underwent, to different extents, adaptive selection as well as purifying selection.

COVID19 is posing threat cosmopolitically encompassing more than 200 countries and making threat to entire population globally as pandemic. The cats, dogs and bovine are at threat which are close partner to human population. The veterinarians specially practicing are at risk when they encounter the sick animals. This study focus to the Pakistani veterinarian where animal population is under estimated or sometimes census is not performed. As Current population of domestic animals in Pakistan consist of 23.34 million buffaloes, 22.42 million cattle, 24.24 million sheep, 49.14 million goats, and with a huge population of dogs and cats without official census which may pose a threat to innocent population and even more a practicing veterinarian and veterinary paramedics are more at threat, if god forbids. There is need for further investigation its role and zoonotic perspective. Regarding practicing field veterinarians fighting without weapons against mass destructing pathogen and making vulnerability on wide.

Incidence of Gastrointestinal Parasites in Zebu and N’dama Breeds from Cattle Ranches in Jos Plateau, Nigeria

The incidence of gastrointestinal parasites in Zebu and N’dama breeds known to be trypanotolerant cattle from some ranches in Jos Plateau was investigated at the peak of the raining season, August to October, 2018. A total of 150 cattle from some Ranches in Jos Plateau were randomly selected for faecal sampling out of which 85 were Zebu (Bos indicus) and 65 were N’dama (Bos taurus) breeds respectively. The samples were analyzed for ova of gastrointestinal parasites using the simple flotation and sedimentation techniques and the results were interesting. The strongyloids, Oesophagostomumradiatum had the highest incidence of 4.7% followed by the Trichostrongylus species with 4%. Fasciolagigantica and Haemonchusplacei both were 3.3%. Then Paramphistomumcervi 2%; bovine hook worm, Bunostomumphlebotomum 1.3%, Taenia saginata1.3%, Schistosoma bovis 1.3% and Nematodirusspathiger 0.7%. The incidence of nematodes, cestodes and trematodes in this study in the raining season was significant (p<0.05) and could pose production and economic threat to institutional farms and ranches in the development of livestock and dairy industries in Nigeria. There should be a strong policy on the control of gastrointestinal parasites in the country for farmers to benefit from their contribution in ensuring food security.

The Raw pH in Plants: A Multifaceted Parameter

The measurement of the in vivo raw pH of vegetative organs is a unusual way obtaining plant knowledge. The authenticity of the pH parameter of the leaf and its independence from soil pH has already been highlighted. In the present work we observe how and to what extent water-temperature mechanisms as well as bio-fertilizers inocula can affect the raw pH and how great the biodiversity is in plants. A trial with Arabidopsis thaliana in a phytotrone has shown that, in the dark, the raw pH did not change from +18 to +35 °C (b = -0.0027 N.S.), while in the light, the regression coefficients were significant and negative, and the acidification in the leaves progressed from high (-0.0097) to normal (-0.0127) and then to low (-0.0370) water level. We have confirmed that warming induces a decrease of raw petiole pH of -0.070 pH C°-1 in grapevine leaves. In accordance with water-temperature mechanisms, the raw pH in grapevines has been found to be significantly higher in well-watered plants (pH = 4.29) than in stressed ones (4.12), with a pH decay of -3.9%. On the other hand, an average reduction of 0.10 units of raw pH would signal an increase in water stress of about -0.59 Mpa. Among the phenomena that can influence the raw pH, we have outlined three biotic factors: i) acidification as a result of a symbiotic farming fertilization i.e through the use of mycorrhizal and microbial fertilizers, with an average decay of around -3%, as a probable signature of symbiosis; ii) an “acida plantarum natura” scenario over 49 species, ranging from pH 3.06 to 6.38 ; iii) a strong (R2= 0.9) inverse polynomial pseudo-relationship of the number of fungicide sprays on the raw pH in a set of 15 species. It is suggested that this simple new multifaceted parameter can deserve interest.

Dream theory from the perspective of Islam

Background and Objective: Sleep as a sign of the God wisdom and power, is the agent of stability and tranquility. Sleep has a spiritual dimension. Sincere dreams are the continuation of prophecy and guidance of the God. This study aimed to explain the dream theory from the perspective of Islam. Materials and Methods: This evolutionary study has focused on these questions: “what is the process of dreaming in Islam? Can people control their dreams”? Islamic evidences were taken from the verses and narrations related to the words “soul, sleep, dream and death”, using al-Mu’ayyim, authentic Shi’a commentary books, Bihar al-Anwar, Kofi principles, the Book of Resurrection and life after death. The religious and scientific evidences were analyzed based on the Walker and Event content analysis method. Dream theory was extracted from religious evidence. Results: Based on "revelation epistemology" during sleep, the ‘template of a higher universe’ travels to the unseen world and divine guidance can be provided. People with spiritual health) (owners of the Sound heart) can control the type of their dream, and solve problems by the guidance of God. In the perspective of Islam, there are three types of dreams: sometimes is divine guidance and glory from God, sometimes is sorrow from the devil, and sometimes are conflicts of daily living or past events. Conclusion: Regarding the spiritual aspect of sleep and its deep impact on physical and mental health, it is essential for medical staff to become familiar with dream theory from the perspective of Islam. They should explain to patients and family members the importance and impact of sleep on the acceleration of tissue repair and the treatment of anxiety and depression. To those who are willing to enjoy the divine guidance through the dream, they can teach the Islamic method of dream control.

First Evidences of Epithelial-Mesenchymal Transition and Cancer Stem-Cell Phenotype Acquisition in Dermo-Epidermal Junction of BPV-Infected Neoplasms

Introduction Bovine papillomavirus (BPV) is the etiological agent of bovine papillomatosis, infectious and neoplastic disease, characterized by the presence of multiple papillomas that can regress spontaneously or to persist and progress to malignancies when in association with environmental cofactors. Although recognized that the BPV can induce DNA damages, the viral role following cancer initiation remains unresolved. Based on this, we stablished cell lines derived from cutaneous papilloma, fibropapilloma and esophageal carcinoma to study the BPV action on epithelial-mesenchymal transition (EMT). Our results showed strong evidences that the virus action can contribute to EMT and, therefore, metastasis. Aim In this study, we analyzed the expression levels of the EMT markers (cytokeratin 10, STAT3 Y705, Oct-3/4 and vimentin) in paraffin-embed samples, using the same tissues that originated the cell lines previous studied, aiming to validate the results observed using cell lines. Material and Methods Expression levels of these markers was analyzed by immunohistochemistry and the collagen composision by Picrosirius red staining. Results We verified an overexpression of these markers in fibroblastoid cells present into the epidermis and ketarinocyte-like cells into the dermis present in dermo-epidermal junction. These data reinforce our previous results using cell cultures, validating both systems (cell culture and paraffin-embed tissues) as useful models to study the natural history of BPV-infected lesions. Conclusion Altogether, the results from these systems indicate that the BPV promote the cancer progression and metastasis through the transdifferentiation of an epithelial to mesenchymal cells (EMT).

Identification and Expression Analysis of Fragaria Vesca MLO Genes Involved in Interaction with Powdery Mildew (Podosphaera Aphanis)

Strawberry powdery mildew, caused by Podosphaeraaphanis is a major fungal disease that affects strawberry yield and quality. In the model plant species Arabidopsis and the crop plants barley, tomato and pea, the Mildew resistance locus O (MLO) proteins have been found to be required for powdery mildew susceptibility. The present study, based on the sequence of a wild plum (Prunus americana) MLO protein, identified 16 MLO genes within the genome of woodland strawberry, Fragaria vesca and examined their expression pattern in response to powdery mildew infection in three diploid strawberry cultivars. Phylogenetic analysis showed that the FvMLO genes can be classified into six clades. Four FvMLO genes were grouped into clade III, which comprises MLO genes from Arabidopsis, tomato and grapevine that mediate powdery mildew susceptibility. A RNA-seq analysis of two diploid strawberry cultivars, F. vescassp. vesca accession Hawaii 4 (HW) and F. vesca f. semperflorens line “Yellow Wonder 5AF7” (YW) at 1 d (1 DAI) and 8 d (8 DAI) after infection showed the expression of 12 out of the 16 FvMLO genes. The comparison of Fragments Per Kilobase of transcript per Million mapped reads (FPKM values) detected by RNA-seq and expression values of qRT-PCR for FvMLO genes showed substantial agreement. The FvMLO3 gene, which was grouped in clade III and orthologous to the Arabidopsis,tomato and grapevine genes, was highly expressed in YW compared to other FvMLO genes across varieties. The results showed that FvMLO genes can be used as potential candidates to engineer powdery mildew resistance in strawberry based on MLO suppression or genome editing.

Assessment of the Immune Response Induced in Neonatal Calves by Vaccination with Mycobacterium Bovis BCG Phipps Under Field Conditions

Immunization of cattle with the bacillus Calmette-Guérin (BCG) vaccine, especially neonates, induces protection against Mycobacterium bovis (M. bovis) and has been proposed as a strategy for bovine tuberculosis (bTB) control. Therefore, the aim of this study was to evaluate the immune response induced under field conditions in neonatal calves vaccinated with BCG Phipps, a strain that has rarely been evaluated in the bovine population, using interferon (IFN)-γ and tuberculin tests, flow cytometry and enzyme-linked immunosorbent assay. Two groups (vaccinated and control) of 5 calves were monitored for 12 weeks, and increases in the in vitro IFN-γ production, the percentage of cluster of differentiation (CD)8+ T cells and the activation levels of CD4+ and CD8+ T cells were observed 3 to 4 weeks post-vaccination. Bovine purified protein derivative-specific IFN-γ production was increased about 4.8- and 5.5-fold in vaccinated animals compared to non-vaccinated animals 3 and 4 weeks post-vaccination respectively. CD8+ T cells of the vaccinated group were increased 1.6-, 1.5- and 1.6-fold at weeks 2, 3 and 4 respectively. Levels of activation were 1.7- and 1.9-fold higher for CD4+ T cells and 2.3- and 1.8-fold higher for CD8+ T cells in the vaccinated group at weeks 3 and 4 respectively in response to M. bovis antigens. However, no animals (vaccinated or control) showed positive results for the single intradermal comparative tuberculin test (SICTT). Therefore, our results indicate that vaccination with M. bovis BCG Phipps strain stimulated peripheral blood T cell activity and induced a cell-mediated immune response. In addition, vaccination did not interfere with the SICTT, as previously reported, which indicates that this vaccine could be successfully applied in bTB control campaigns.

Family with Sequence Similarity 213, Member A is Associated with the Fertility Selection Index in Holsteins

Female fertility is an economically important trait in the dairy industry, and the fertility selection index has been developed as a method of including female fertility in the breeding goals of this industry. This index considers a combination of factors, including days open, number of inseminations per lactation, success after first insemination, and pregnancy within 70 d, 90 d, and 110 d after parity. Based on a genome-wide association study of the fertility selection index using 442 Holsteins, we found that the index is influenced by a variation in the thioredoxin fold region of the family with the sequence similarity 213, member A (FAM213A) protein. FAM213A is a CXXC motif-containing peroxiredoxin 2-like protein that regulates cellular redox status. A replacement of isoleucine with valine in FAM213A was associated with poor fertility in cows. The overexpression of FAM213AVal in bovine endometrial epithelial cells reduced reactive oxygen species to a lesser extent relative to the overexpression of FAM213AIle and caused a decrease in cyclooxygenase-2 expression. Downregulation of cyclooxygenase-2 led to a decline in prostaglandin E2, which is critical for implantation because it protects the conceptus from the maternal immune system. Cows with FAM213AVal showed lower levels of prostaglandin E2 than did cows with FAM213AIle, suggesting that cows with FAM213AVal are less fertile than cows with FAM213AIle because of their reduced uterine environment. Thus, the present study found that FAM213A unexpectedly modulates female fertility in cattle.

Composite Photopolymerization: Temperature Increase According to Light Source and Dentin Thickness

When a composite resin restoration is photopolymerized, a considerable amount of heat can be produced, potentially causing pulpal injury. Objective: Evaluate the influence of the type of light source and remaining dentin thickness on the temperature increase in the pulp chamber while curing composite resin restorations. Material and Methods: Ninety-six tooth fragments obtained from bovine incisors were divided into groups/subgroups (n=12), according to the light source (LED and halogen light) and remaining dentin thickness (3.5, 3, 2, and 1mm). Class I cavities were prepared and restored with a composite resin. A temperature increase was obtained during photopolymerization of the adhesive and each composite increment. Data were analyzed using ANOVA and Fisher’s Test (α=5%). Results: LED promoted higher temperature increments when compared with the halogen light. Temperature levels were the lowest for 3.5mm-thick and the highest for 1mm-thick remaining dentin. Levels registered during the photopolymerization of each composite increment were superior for LED. Conclusions: Both light sources result in temperature increases above 5.5°C. Additionally, the remaining dentin thickness of 1mm promoted the largest temperature increase.
