Overview
Alzheimer's disease is a progressive neurodegenerative disorder and the most common cause of dementia, characterized pathologically by extracellular amyloid-beta plaques, intracellular neurofibrillary tangles of hyperphosphorylated tau, synaptic loss, and neuroinflammation, leading to decline in memory, language, and executive function. Onset is typically insidious in later life, and although no curative therapy exists, intervention can modify symptoms and disease trajectory, making early detection and characterization important research priorities. Work in this area spans candidate biomarkers and imaging approaches, including optical coherence tomography of the retina for early prediction and the relationship of basal serum cortisol and medial temporal lobe atrophy to mild cognitive impairment and Alzheimer's disease, alongside emerging molecular markers such as circular RNAs and plasma TREM2. Therapeutic exploration includes vagus nerve stimulation, while caregiver-focused research distinguishes genuine knowledge gaps from misconceptions among those supporting affected individuals. Behavioral and sleep dimensions are also addressed, including the neurobiology distinguishing aggression from agitation in dementia and non-pharmacological interventions for disrupted sleep in moderate-to-severe dementia. The journal publishes peer-reviewed studies on biomarkers, imaging, therapeutics, caregiving, and the behavioral and sleep manifestations of Alzheimer's disease and related dementias, addressing early diagnosis and management across the disease course.
